Precautions for freeze-drying
Freeze dried samples should be pre frozen to a solid state to avoid direct freeze-drying of liquids. Avoid containing high concentrations of organic solvents such as ethanol and acetone, which may corrode equipment or pose a risk of explosion. If it contains volatile components, they need to be removed in advance or protective measures should be taken. The packaging thickness should be uniform (usually not exceeding 1 cm) to avoid incomplete drying caused by excessive thickness. Use low-temperature resistant containers (such as specialized freeze-drying bottles) to prevent glassware from breaking due to low temperatures. The bottle mouth is covered with a breathable sealing film to prevent contamination and allow water vapor to escape. Ensure that the sample is completely frozen (at a temperature below the eutectic point, typically below -40 ℃). Insufficient pre freezing may cause the sample to melt or bubble during the drying process. The loading capacity should not exceed the maximum capacity of the freeze dryer to avoid overloading of the cold trap (generally ≤ 70% of the surface area of the cold trap). Leave gaps between sample bottles to ensure even circulation of cold air.
